The plot of Sookshmadarshini centres on Priyadarshini, also known as Priya (Nazriya Nazim Fahadh), a stay-at-home mom who lives in a tight-knit suburban community with her husband and daughter.
When someone hits Priya's car's bumper at the beginning of the movie, she starts to wonder who did it.In her day-to-day life, Priyadarshini is just a regular woman in dire need of employment.
Priyadarshini is renowned for her detective abilities, so when Manuel (played by Basil Joseph) and his mother, Grace, move into their suburban neighbourhood, her life takes an intriguing turn.
The neighborhood's long-time residents, Manuel and Grace, who the former owned a bakery in the city, moved out a few years ago. Manuel presents a positive front right away, concealing his actual motivations.
Priya appears to view him as a dubious individual who is making a concerted effort to conceal a secret, even though everyone finds him impressive. Meanwhile, it is disclosed that Manuel, his uncle Roy, and his cousin Dr. John are preparing to commit a terrible crime.
Furthermore, it is reported that Grace, Manuel's mother, has Alzheimer's disease and is not aware of what is going on in the house.
Because it's their wedding anniversary, Priya and her husband decide to throw a party during this time. Manuel's mother disappears when his house catches fire during the party.
Diana, Grace's daughter, returns home after Grace disappears, but she leaves for New Zealand because the investigation has not yielded any results.
Priyadarshini becomes suspicious as a result of the circumstances and timing, but one evening, she sees Grace. But nobody takes her seriously. Priya chooses to use her detective abilities to solve the mystery because she is tired of the stressful circumstances.
Priyadarshini tells her group of friends in the neighbourhood that she believes she saw Grace, and they all work together to solve the crime.
Manuel accuses Priya of bothering him while he is coping with his mother's disappearance on several occasions. The true twist, though, occurs when Grace goes back to her house and is still unable to talk about what happened to her.
Priyadarshini makes one last effort to speak with Grace, but Manuel cuts her off once more. Priya contacts Grace's daughter Diana, who is still dubious about everything.

How does Priyadarshini learn the truth, and what happened to Grace?
Dr. John tries to do something nefarious with medical equipment inside Manuel's house while Priyadarshini tries to get information from Diana.
Diana abruptly stops answering her texts, so Priya surveils her social media accounts. It is soon discovered that Diana is a lesbian who lives with Aditi Thyagarajan, a Tamil woman, with whom she planned to adopt a child in New Zealand.
When Priya talks to Aditi, she finds out that she is in India looking for Diana and that Manuel has volunteered to meet her at the airport.
Priyadarshini acknowledges that her suspicions were correct, but that Diana, not Grace, was the real victim. Priya locates them and saves Aditi as Manuel attempts to kidnap and murder her.
By reliving the events, it becomes clear that Grace was the real offender and that she is not afflicted with Alzheimer's. She believes Grace has disappeared, so she, her brother, and her son decide to kill Diana after being disappointed that her daughter is a lesbian.
It is then disclosed that Manuel was the one who texted Priya under the guise of Diana in order to deceive her. All of them are arrested at the end of the film, shocking everyone except Priyadarshini.
In a last callback, it is revealed that Manuel was responsible for Priya's car being struck at the start of the movie, suggesting a thematic link between their lives.
